A Fur-regulated type VI secretion system contributes to oxidative stress resistance and virulence in Yersinia pseudotuberculosis

چکیده

Abstract The type VI secretion system (T6SS) is a widespread protein apparatus deployed by many Gram-negative bacterial species to interact with competitor bacteria, host organisms, and the environment. Yersinia pseudotuberculosis T6SS4 was recently reported be involved in manganese acquisition; however, underlying regulatory mechanism still remains unclear. In this study, we discovered that regulated ferric uptake regulator (Fur) response ions (Mn 2+ ), negative regulation of Fur proceeded specifically recognizing promoter region Y. . Furthermore, induced low Mn oxidative stress conditions via Fur, acting as -responsive transcriptional maintain intracellular homeostasis, which plays important role transport for survival under stress. Our results provide evidence can enhance resistance virulence This study provides new insights into -dependent expands our knowledge mechanisms functions T6SS from

Expression of a Yersinia pseudotuberculosis Type VI Secretion System Is Responsive to Envelope Stresses through the OmpR Transcriptional Activator

The Type VI secretion system (T6SS) is a macromolecular complex widespread in Gram-negative bacteria. Although several T6SS are required for virulence towards host models, most are necessary to eliminate competitor bacteria. Other functions, such as resistance to amoeba predation, biofilm formation or adaptation to environmental conditions have also been reported. This multitude of functions is...

IscR Is Essential for Yersinia pseudotuberculosis Type III Secretion and Virulence

Type III secretion systems (T3SS) are essential for virulence in dozens of pathogens, but are not required for growth outside the host. Therefore, the T3SS of many bacterial species are under tight regulatory control. To increase our understanding of the molecular mechanisms behind T3SS regulation, we performed a transposon screen to identify genes important for T3SS function in the food-borne ...

Innate Immune Recognition of Yersinia pseudotuberculosis Type III Secretion

Specialized protein translocation systems are used by many bacterial pathogens to deliver effector proteins into host cells that interfere with normal cellular functions. How the host immune system recognizes and responds to this intrusive event is not understood. To address these questions, we determined the mammalian cellular response to the virulence-associated type III secretion system (T3S...

Extracytoplasmic-stress-responsive pathways modulate type III secretion in Yersinia pseudotuberculosis.

Three signal transduction pathways, the two-component systems CpxRA and BaeSR and the alternative sigma factor sigma(E), respond to extracytoplasmic stress that facilitates bacterial adaptation to changing environments. At least the CpxRA and sigma(E) pathways control the production of protein-folding and degradation factors that counter the effects of protein misfolding in the periplasm. This ...
